This study investigated the greenhouse gas emissions of amusement parks in Taiwan. The energy use of 26 parks evenly distributed throughout Taiwan was audited. An average of 583,000 people was found to visit each site annually. In addition, average annual energy consumption of 7699 MW h per site was observed, translating into 4096 t of carbon emissions per year. Every visitor generated an average of US$29.4 in revenue and 7.4 kg-CO2eq. The parks offering luxury services, such as 4-or 5-star accommodation and luxury food products, or many large-scale motorized water- or ground-based attractions consumed more energy than did ecology- and education-oriented parks. We categorized the parks into five categories, and found significant discrepancies among them in energy use. Two models for predicting annual carbon emissions and average carbon emissions per visit were developed, with the adjusted coefficient of determination being 0.934 and 0.792. This paper concludes by proposing several guidelines to assist park operators in reducing carbon emissions and improving energy efficiency. © 2017 Elsevier Ltd

Sinularin is an active compound isolated from the cultured soft coral Sinularia flexibilis. In this study, we investigated the effects of sinularin on two human gastric cancer cell lines, AGS and NCI-N87. Our results demonstrated that sinularin suppressed the proliferation of gastric cancer cells in a dose-dependent manner and induced apoptosis. In addition, the loss of mitochondrial membrane potential, the release of cytochrome C, the activation of Bax, Bad and caspase-3/9, and the suppression of p-Bad, Bcl-xL and Bcl-2 were observed in the cells treated with sinularin. This finding suggests that sinularin-induced apoptosis is associated with mitochondria-mediated apoptosis and occurs through caspase-dependent pathways. Furthermore, sinularin inhibited the phosphoinositol 3-kinase/Akt/mechanistic target of the rapamycin signaling pathway. Taken together, our results show that sinularin-induced apoptosis is mediated by activation of the caspase cascade and mitochondrial dysfunction. Our findings suggest that sinularin merits further evaluation as a chemotherapeutic agent for human gastric cancer. © 2016 by the authors; licensee MDPI.

Methylglyoxal (MG) is a reactive dicarbonyl compound that is produced endogenously from glycolytic intermediates and via gluconeogenesis. Elevated MG levels in diabetes patients are believed to cause diabetic complications. MG-induced crosslinking products from the covalent binding of DNA or protein alone or together could be relevant to carcinogenesis and multiple complications in diabetes. However, the mechanisms governing DNA crosslink formation by MG are unclear. We investigated whether MG could induce DNA crosslinks in human ECV304 cells and the possible mechanism of this action. The level of DNA crosslinks and reactive oxygen species production were assayed by a modified alkaline Comet assay and a 2',7' dichlorofluorescin diacetate (DCHF-DA) assay, respectively. MG caused a time- and dose-dependent increase in DNA crosslinks and a dose-dependent increase in protein carbonylation in ECV304 cells. Addition of 2. mM MG resulted in a transient increase in protein carbonylation, and this increase peaked within 2. h and then rapidly decreased. Most notably, MG did not cause significantly enhanced ROS generation in ECV304 cells. Co-treatment with carbonyl-scavenging drugs, such as aminoguanidine, N-acetyl-. l-cysteine, and glutathione, significantly inhibited the formation of DNA crosslinks by MG, whereas co-treatment with the antioxidant ascorbic acid did not. In conclusion, our results imply that MG induces DNA crosslink formation in ECV304 cells via a reactive oxygen species-independent protein carbonylation pathway. Our findings also suggest that non-toxic aminothiol antioxidants with carbonyl scavenging capabilities are potential therapeutic agent for MG-related diseases, such as diabetes and neurodegeneration. Furthermore, our findings also imply that DNA nonbinding proteins, bovine serum albumin might be able to crosslink calf thymus DNA in the presence of MG. © 2013 Elsevier Ltd.

A new spatane diterpenoid, leptoclalin A (1), along with two previously reported known norcembranoid diterpenes (2 and 3), were isolated from a cultured soft coral Sinularia leptoclados. The structures were determined by extensive spectroscopic analyses and by comparison with the spectral data of related known compounds. Metabolite 1 is rarely found in spatane skeletons reported from soft corals. In addition, compound 1 exhibited weak cytotoxicity towards human tumor cell lines T-47 D and K-562. © 2013 by the authors.

The effects of shikonin on gastric cancer cells were investigated in this study. Exposure to shikonin reduced the viability of gastric cancer cells in a time- and dose-dependent manner. However, apoptosis was not observed in gastric cancer cell treatment with different concentrations of shikonin for 6 h. By contrast, treatment with shikonin for 24 h significantly induced apoptosis, as evidenced by the results of TUNEL assay and flow cytometry analysis in proportion to the concentration. Disruption of the mitochondrial membrane potential was observed in gastric cancer cells that were treated with shikonin for 6 and 24 h. Pretreatment with necrostatin-1 recovered cell death and mitochondrial membrane potential in the 6 h shikonin treatment, but not in the 24 h shikonin treatment. Western blot results reveal enhanced p38 phosphorylation, downregulated AKT phosphorylation, and increased caspase3 and PARP cleavage in cells that were treated with shikonin for 24 h, but not in cells treated for 6 h. Shikonin also triggered reactive oxygen species (ROS) generation both in the 6 and 24 h treatments. Pretreatment with N-acetylcysteine blocked shikonin-induced cell death. In summary, our findings suggest that shikonin, which may function as a promising agent in the treatment of gastric cancers, sequentially triggered necrosis or apoptosis through ROS generation in gastric cancer cells. © 2014 Elsevier Ireland Ltd. This study deals with the emerging problem of jointly determining the optimal retail price, the replenishment cycle, and the number of shipments for exponentially deteriorating items under conditions of channel coordination, joint replenishment program, and pricing policy. In a one-manufacturer and multi-retailer channel setting, it is assumed that the retailers' demands are price-dependent and can be influenced by time. Two profit-maximization models including the non-integrated policy and the integrated policy were formulated with the aim of maximizing the channel-wide profit. In addition, this study demonstrated the optimal properties of the models, developed a search algorithm to obtain the optimal solutions, and offered structured and quantitative insights into the interplay between the proposed models through a numerical study. © 2009 Springer-Verlag London Limited.

Pressure sore reconstruction is always a challenge for plastic surgeons due to its high recurrence rate. In addition to the myocutaneous flap, the perforator-based fasciocutaneous flap has become a new entity used for pressure sore reconstruction. This study presents a series of 26 perforator-based fasciocutaneous flaps for pressure sore reconstruction, with good outcomes in 21 patients from July 2008 to April 2011. The flaps were advanced, transposed, or rotated to obliterate the defects. Twenty of 26 flaps healed uneventfully without complication. One patient had a flap that totally necrosed, one had partial flap necrosis (flap rotated 180°in the above two cases), one had infection and healed by a secondary flap, one had minor wound dehiscence, one died of pneumonia 1 week postoperatively, and recurrence developed in one patient. The perforator-based fasciocutaneous flap is a reliable method and produced good results in this series. These flaps are well vascularised, have enough soft tissue bulk, and have a high degree of mobilisation freedom. © 2012 Informa Healthcare.

Background: Congenital dislocation of the knee (CDK) is a very rare condition that comprises a spectrum of deformities from subluxation to complete dislocation. The incidence of CDK is estimated at 1 per 100,000 live births, which is 1% of the incidence of developmental dysplasia of the hip (DDH). Moreover, 40-100% of patients with CDK have additional musculoskeletal anomalies, the most common being DDH and clubfoot. In general, the diagnosis is established immediately after birth according to the position of the knee recurvatum. Treatment with conservative methods at an early stage is most likely to yield successful results. We report here successful treatment of a series of CDK patients with early reduction. Methods: From July 1990 to June 2007, 19 patients with CDK (affecting 25 knees) were treated with early reduction. Of these, 6 knees had dislocation, and 19 had subluxation. Since 1990, treatment has been guided by a protocol that considers patient age and the severity of the condition. In patients examined within 24 hours of birth, early, direct reduction under gentle, persistent manual traction was attempted. Birth history and perinatal course were obtained from medical records. Associated musculoskeletal anomalies were observed and treated after reduction of the knee joint. A Pavlik harness was used for at least 4 months in the concomitant treatment of DDH and CDK. Knee function was graded as excellent, good, fair, or poor. Radiographs were used to assess DDH during follow-up. Results: After an average follow-up duration of 4.3 years, 18 patients showed an excellent or good outcome. One patient, whose knee could not be reduced, had severe multiple anomalies and died 16 days after birth. Fifteen of the nineteen patients had associated musculoskeletal anomalies, including DDH and foot deformity. Two cases of residual hip dysplasia after Pavlik harness application required an acetabular osteotomy. Conclusions: For CDK patients, early and direct closed reduction within 24 hours of birth affords outcomes graded as either excellent or good.

Background: Maize contaminated with mycotoxin fumonisin B1 poses a global threat to agricultural production. In this study, polyclonal antibodies (pAb) specific to fumonisin B1 were generated from rabbits immunised with fumonisin B1-keyhole limpet haemocyanin (KLH). These antibodies were used to establish a sensitive competitive direct enzyme-linked immunosorbent assay (cdELISA) and gold nanoparticle immunochromatographic strip for detecting fumonisin B1 levels in maize-based foods and feeds. Results: In cdELISA, fumonisins B1, B2 and B3 at concentrations of 0.42, 0.58 and 81.5 ng mL-1 respectively caused 50% inhibition (IC50) of binding of fumonisin B1-horseradish peroxidase (HRP) to the antibodies. Effective on-site detection of fumonisin B1 was achieved by developing a rapid and sensitive pAb-based gold nanoparticle immunochromatographic strip. This strip had a detection limit of 5 ng mL-1 for fumonisin B1 in maize-based samples. Additionally, the whole analytical process could be completed within 10 min. Close examination of 15 maize-based samples by cdELISA revealed that 11 were fumonisin-positive, with a mean concentration of 435 ± 20.1 ng g-1. These results correlated well with those obtained by immunochromatographic strip.Conclusion: Both cdELISA and immunochromatographic strip methods established in this study are sensitive for rapid detection of fumonisins in agricultural commodities. © 2010 Society of Chemical Industry.

Taiwan is famous for its personal computer (PC) products, and this success demonstrates that Taiwanese manufacturers have effective strategies in managing supply chains of PC-related products. Among various strategies of supply chain management, achieving leagility is the main goal. The purpose of this study is to illustrate how a PC original equipment manufacturer (OEM) in Taiwan achieves leagility through reengineering its supply chain. The case study shows how the company adjusts its production processes from build-to-order (BTO) to configuration-to-order (CTO) to achieve leagility.
